Codeine piperidine
Just wondering if a 300mg codeine and black pepper mix would do anything to add to the experience as they contain cyp2d6 enzyme inducers. Please help need an answer asap.

I've done some research into this and I wouldn't recommend it. Piperine alters both the CYP450 isozymes and the activity of enzymes involved in glucuronide/sulfate conjugation. If you've used a paracetamol(acetaminophen) containing product to obtain your codeine the end result will be increased toxicity from the paracetamol due to interference with the paracetamol detoxification pathway. I haven't looked into aspirin/ibuprofen detoxification pathways but there's a possibility that that would be risky too.
If you have pure codeine you're safer, but messing around with P-GP inhibitors sounds dangerous too - they keep many toxic substances out of the brain, so if you did use piperine you'd have to fast and refrain from smoking to be on the safe side, and even then you couldn't be sure that endogenous compounds weren't getting into the brain and causing meyhem. I'd say, don't do it. malsat added 1 Minutes and 49 Seconds later... The codeine is a syrup with pseudo and triprolidine. Black pepper has piperine by mass 5-9%.

Well tripolodine doesn't appear to be a p-gp substrate and I don't know if pseudo is, but if it is then you'll be getting a nasty whack of stimulation if you do it. You'll have to look into the detox. pathways of both and see if the enzymes listed in my bluelight post are involved so that you don't end up with toxic metabolites being produced or overdosing on either due to inhibited clearance. The reason I say I wouldn't know how much pepper to take is that I don't know what amount of piperine would lead to acceptable p-gp inhibition as I haven't seen any studies on the topic.
If you do decide to do it, I'd guess that you should start off with the amount of black pepper a person would put on a meal, wait a while before dosing... and then based on the effectiveness of that you could increase the pepper dose by a reasonable amount next time and repeat. But just remember if you do go ahead with it, it's your body and you're playing guinea pig! Personally I decided against it when I was thinking about it. |
