Had 1mg of Bremelanotide (PT-141) left over from my last experiment with the stuff. Figured it wouldn’t do anything, but it was just lying around anyway, so I tried it intranasally to see if Palatin’s in-development delivery system could be recreated. It works folks. Intranasally as well as sub-cutaneously — it works. Something that I think I didn’t adequately describe in previous entries is the degree to which this drug does NOT act acutely. It takes hours and hours (after the initial flushing effect) to experience its pro-sexual effects. Perhaps 3-4 hours after dosing, I fell asleep, having not yet experienced the pro-sexual effects. But this morning I would have broke the rigiscan — that strange contraption they use in clinical trials to measure the effects of “male research” drugs — had I been subjected to it. Another thing to emphasize is the utter randomness of the erections. Viagra works very simply: viagra + sexual stimulation = bonerville. PT-141 is more like puberty. Random erections at strange times and the like. Vaguely uncomfortable.




IS BREMELANOTIDE (PT-141) THE ANSWER?

That said, a drug I’ve currently been messing around with called Bremelanotide (previously called PT-141) has been touted by some as the female Viagra. In rats, at least, Bremelanotide increases “female sexual solicitation behavior,” an exciting (so to speak) finding to say the least. Then again, if rat behavior perfectly correlated with that of humanity, obesity would be a thing of the past: diet drugs frequently reduce the body weight of lab rats by 40% or so! But Bremelanotide is notable for other reasons: as opposed to Viagra and its fellow PDE-5 inhibiting peers (Levitra, Cialis), Bremelanotide acts on the brain, not the vasculature of the body, striking the “melanocortin receptors” in the hypothalamus. I’d go on if I thought you’d listen…

So, does Bremelanotide work? Is it the female viagra, or is it bunk junk?

Alcibiades’ bold prediction is that this stuff will work — not for every female — but that it will exert a pro-sexual effect on a considerable subset of the female population. I base this on personal experience, though the careful reader will note that I have been endowed with the Instrument of Adam, not Plato’s Cave.

My own personal experience with the drug was suggestive. And the thing about Bremelanotide is that its effects are very difficult to mistake for Lady Placebo. Acutely, it flushes the face like — well, a lot like the head of an engorged…tomato. Quite a while after the initial dosing after, often 3-6 hours later, you become aware of a “pressure” in the loins. It’s something like an internal tickle down there. And, behaviorially, you just end up grabbing your ding dong more. You just sort of need to. It’s like a compulsion or something.

Now unfortunately, not everyone is as enthusiastic about “introspective research” of unapproved pharmaceuticals as I, so I cannot confirm that these effects are the same in women. Still, the more-than-just-blood-flow effects I experienced lead me to believe that this stuff may well do something for the fairer sex.

Bremelanotide, if approved, will not be available for another few years. For the FDA, like a hot date, likes to make us wait. If in the meantime the anticipation is killing you (and you are a post-menopausal women), you might try the testosterone patch, which seems to be useful for some — though the potential for masculinizing effects scare a lot of folks off. (Probably rightly.) Perhaps the most important thing to do if you are a female suffering from sexual dysfunction is to make sure you make sure you aren’t already on something that could be affecting your libido: SSRIs, for instance, notoriously affect sexual drive and functioning. Different birth control pills insofar as they effect hormonal systems can also have dampening (or paradoxically, enhancing) effects on libido, so mess around with those. Finally, a caring, devoted partner is especially important for women, whereas selfish-gene-programmed, spread-your-seed men tend to be better equipped to handle multiple, changing partners.

And that’s all I have to say about that.



Brem is for Real

First off, I’m sick of typing out Bremelanotide (PT-141). From now on it’s Bremelanotide. Scratch that, it’s “Brem.”

Injecting Brem 4mg sub-q was an interesting experience. First off, I’m evidently pretty rusty from my days of slamming steroids on a daily basis. In those days, when I was sticking girthy 23″ gauge needles 2 inches into my ass cheek, I would have laughed at my present-self experiencing a profound sympathetic response to the act of slipping a diminutive insulin needle under the skin.

(Note that I wasn’t about to waste a prohibitively expensive compound by going the intranasal route. As alluring as it is to have libido in an easy-to-inhale form. Great marketing by the way, Palatin.)

So I constituted the peptide (put the powder into bacteriostatic water), prepared a 4mg-in-1ml dose, and injected it into the subcutaneous fat layer that remains atop my stomach despite prodigious cardio on my part!

20 minutes later, my face turned way-the-fuck red. Mars-rock red. Uneven blemishes and former acne became very visible, lit up seemingly from within like Christmas lights. I definitely was experiencing a headache. All in all, I didn’t feel so good.

No spontaneous erections to speak of. No increased desire for my fellow classmates, or, for that matter, the scantily- or not-so-clad women I “tested” myself with on the internet.

When the flushing and headache diminished perhaps an hour later, I was ready to dismiss this drug as more drug company hype. Yeah, blood flow was effected at some level, but a “designer aphrodisiac” Brem was not.

Approximately 3 hours after dosing, I found out how wrong my initial assessment was.

The most peculiar thing: a sort of tingling, pressing, weighty feeling at the base — a bit deeper — of my Nimbus 2000. I won’t say that I was about to jump the maid, as that might be ad hoc ascribing something psychological to what I felt, but again, there was a “squeeze me” feeling thumping in my man-unit. I was very much reminded of the feelings I had had when going through that strange part of life when hair grows out of our armpits. That part of your life where despite all risks you engage in a bit of self-inflicted Ultraviolence even though your mom’s in the next room over.

The effects of Brem were immensely prominent until I went to sleep (6 hours later?) hours following the inject. Additionally, random erections, uncomfortable jeans-hugging stiffies, and especially that ever-present “need to satiate” were very noticeable for the next few days. (I suspect the duration of effect is due to large amount used and to the “slow trickle” of active compound from fat stores into the blood stream, though this is just a hypothesis.) Pronounced yawning as well, which jives with Brem’s purported action on the hypothalamus, and its effects on the old pogo stick.

Alcibiades’ Bottom Line: Brem is for real.

Banging Bremelanotide

Ummmm…Bremelanotide (a.k.a. PT-141) most definitely works! The flush reminds me of a woman’s “sex flush.” I even took pics of myself to document the remarkable deep red (tomato-like) color you turn — it is not normal! (No, I’m NOT showing you!) The psycho-physiological effects are distinctively reminiscent of puberty. There is a strange tightening in my pelvic region that I perceive the need to satiate.

Bremelanotide - arrival at the black market?

Bremelanotide, the new "first real aphrodisiac", is not finished yet, for approval of the FDA (will be sometime in 07/08).

The drug was initially used for tanning. And it's chemical makeup goes like this:

The invention relates to a peptide that is a free acid or pharmaceutically acceptable salt thereof that includes the sequence His-Phe-Arg-Trp (SEQ ID NO:1), His-D-Phe-Arg-Trp, homologs of His-Phe-Arg-Trp (SEQ ID NO:1) or homologs of His-D-Phe-Arg-Trp. The peptide is preferably a cyclic peptide, and preferable has a terminal --OH at the carboxyl terminus. In a preferred embodiment, the peptide is Ac-Nle-cyclo(-Asp-His-D-Phe-Arg-Trp-Lys)-OH.


Not sure if that makes sense to anyone, however I've seen it on the black market already. But only at one place..

I'm far from being a chemist, so I wonder is this something which would spread throughout the "underground labs"? Are tanning products manageble to make without big $$$ equiptment

It has been offered to the public for huge amounts of money. The chinese are now producing this and spreading it to internet sources, which are able to do so in a reasonable way. If this is effective like described on the net, this must be worth synthesising.

Bremelanotide Promises to Help Both Men and Women Find That Lost Loving Feeling

What is desire? What triggers it? And how can we make it last?

Some people think oysters, chocolate and ginseng -- among other mythical things --are aphrodisiacs. They are rich in nutrients and give energy, but no one has proven they give you passion.


Even the "little blue pill" Viagra just works on, shall we say, the plumbing -- it keeps the blood flowing in the right direction.

Now there's a drug in the pipeline that its makers say really will restore lost libido.
It's being tested and developed, in part, in a laboratory at Concordia University in Montreal by neuroscientist Jim Pfaus.


Full-time college professor, part-time punk rocker, Pfaus is using rats to test whether the new substance, bremelanotide (pronounced "BREE-ma-LAN-o-tide") -- or PT 141 -- triggers desire.


Rats stand in for humans, because, like people, they're social and they have a similar hypothalamus, the part of the brain that controls desire.


In his lab, Pfaus says he's finding that bremelanotide seems to put rats in the mood.
Given the peptide, female rats, he says, initiate sex four times more often than those who do not receive it.


And bremelanotide's makers are betting it will work the same way on both men and women.
"It brings back your libido," Pfaus said. "It doesn't make it something that it wasn't. It brings it back to where it probably was when you were having good sex."


[h4]From Tanner Enhancer to Sex Driver[/h4] Bremelanotide didn't start out as a drug for sexual dysfunction.


In fact, it was being developed as a tanning enhancer, until researchers noticed interesting side effects in the men involved in clinical trials.

"All treatments for sexual dysfunction, especially erectile dysfunction, have colorful stories about how they started," said Carl Spana, CEO of Palatin Technologies, which holds the patent on bremelanotide. "In this particular one, young college men were given this drug in a Phase 1 study and got spontaneous erections.

Spana hopes his company has stumbled on the holy grail of sexual dysfunction.

And how does bremelanotide work?


Spana says it activates parts of the brain that are involved in regulating normal sexual function.

e says other products on the market, such as Viagra, Cialis and Levitra, "work by affecting peripheral blood flow, [rather than] the central nervous system."
[h4]A First of a Kind for Women[/h4] Palatin Technologies, a biotech company in New Jersey, hopes to have bremelanotide on the market for men within a couple of years -- and for women shortly after that.

If approved, it would be the first drug sold in the United States to specifically target lack of sexual desire in women.


It's for those experiencing serious sexual problems -- women who've gone through menopause or had a hysterectomy, for instance, and lost desire as a result.

Michael Perelman, a New York City sex and marriage therapist, screened women for Palatin's human trials, and has kept an eye on the results.


"Some of the women put it in colorful language that I'm not comfortable using on television," Perelman told ABC News' "Nightline."


"But what was said was that, 'The quality of my orgasm was more the way it used to be, and I like that.' 'I had some thoughts about my husband.' 'I initiated sex with him for the first time even though I've thought about doing it for a long time.' Those were some of the things that they said," Perelman said.

If and when bremelanotide makes it to market, it will be administered as a single-use nasal spray.


That way, it goes straight to the brain and avoids being broken down in the digestive system.


Dr. Annette Shadiack, Director of Research at Palatin, says the results last six hours or eight hours, with the onset as early as 30 minutes.

[h4]Drug Abuse Concerns[/h4] But bremelanotide likely will encounter tough resistance on its way to receiving approval from the Food and Drug Administration.


One concern is abuse. Some young partyers have been known to mix Viagra with Ecstasy for recreational use.

"I think that there'll be enormous sales over the Internet. I think that there'll be sales in bars, on street corners," said sexual psychologist Leonore Tiefer, associate clinical professor of psychiatry at the New York University's School of Medicine. "Certainly when the thing is first approved, I think it will be widely misused."


In 2004, Tiefer helped block FDA approval for Proctor & Gamble's hormone patch Intrinsa, which was being touted as the female Viagra.

Besides the potential for abuse, she also worries about the overall impact of these sorts of drugs on the culture.


In particular, she's worried about the way it will be marketed.


"It raises the bar on performance expectations. So that women, couples, will somehow feel that if arousal, orgasm, desire, is not a regular routine, every day, every week, womb-to-tomb kind of thing that there is something wrong with you," Tiefer said.

"If women do go in with that message, their physician should clearly tell them that this is not what this is for," said Palatin CEO Spana. "It's for women who feel a real lack of desire, real lack of ability to get aroused."


Back in his lab in Montreal, Pfaus says he thinks women should have the option, but he hopes bremelanotide will give women confidence, not doubt.

"Women don't need another thing staring at them from a billboard telling them that they are not good enough," Pfaus said.


"But I think that if the drug is available for people who actually have true desire disorders that are defined by a psychiatrist and a clinical psychologist, then we're talking about a different group of people," he said.


But Pfaus says critics should not be overly concerned about bremelanotide's potency.
"This is not going to make people have orgies in the street," he said.

Among women who attempted sexual intercourse within 24 hours after treatment, significantly more were satisfied with their level of sexual arousal following bremelanotide, compared with placebo (P = 0.0256). It is, after all, the cheap, easy-to-use, hassle-free sex drug everyone's been waiting for. It is nirvana, the magic bullet, the simplest route to quickie sex you've ever known. It could be the greatest thing to happen to sex in 50 years. And also, of course, the worst.

cabergoline isn't a take your clothes off and do me substance for sure though...let's just say it enhances the experience, in swim's (male) experiences. oh, and then there's the dizzyiness to deal with

PT-141 sounds like a fantastic drug. SWIM may indulge in it when he's old and withered. Sounds like a more natural alternative to viagra in terms of its effects. SWIM wonders, are there any known side effects in combining with other drugs-- alcohol, opiates, stimulants?

According to the article I posted above there is no interactions with alcohol. It's unlikely that the FDA will explore interaction with illegal substances, so we'll just have to wait for valiant explorers when PT-141 hits the market.

Just a quick word on this: After a recent personal experiment with a vial of PT-141 and after reading experience reports on another forum from several other people who've tried it, SWIM thinks this drug is being massively over-hyped and doesn't even come close to living up to the potential described in the media reports.

Will update with details on SWIM's experience later...

Couldn't agree more - can't wait 'til it's on the market. Remember men, a women's libido is at a slightly lower level than yours. If this drug is already labelled as cum drug for women the FDA will think that it makes women horny so they will most probably think it could be used as a date-rape drug. Then a few idiots will use it with that purpose and it will be banned instantly.

The drug is obviously a good think, but all the hype around it is bad, very bad.

No offense, Toe, but I think you're attacking a strawman here. No one was implying any sort of sexist sentiment. The researcher expressing the quoted concerns is a woman fearing for other women, not a man trying to oppress females. That being said I whole heartedly agree with you, and I do think there's still a puritanical slant towards sex and drugs in the US. This sentiment might lead to the sort of thinking you seem to be worried about, which is that the drug might be suppressed simply because the idea of using a drug just to increase a woman's (or man's) sexual enjoyment is wrong.

The thing is the concerned woman researcher does have a point. Due to the massive US pharmecutical lobby, and the unique american captalist laws that allow for aggressive drug marketing to the public, it's easy for people to be misled into using perscriptions which are harmful and unncesary. Every new perscription drug is marketed as a non addictive, safe, miracle drug when it's released (oxycontin, anyone?) until it comes out years later that the drug has been vastly overperscribed and causes serious problems.

Not to mention, all "moral" arguments aside, I do think there's something to be said for perfectly normal adults to have sex without the aid of drugs. There are already illegal libido increasing drugs out there, and while using them for the occasional intense sexual encounter is fun, getting into the routine using a certain drug every time you want to have sex can be a bad habit.

A few idiots misusing something doesn't necessarily lead to an instant ban by the FDA. If a big pharmaceutical house has enough R&D money sunk into this, it's going to take a great deal more than a few dummies misusing it for the DEA to override the FDA. Case in point: Viagra, Cialis and Levitra are all still on the market, after several heart-attack related deaths hanging over them and all that was required by the FDA was a mention in the literature.

Fully agree with BadnessNeeds, couldn't agree more! But what's more, since bremelanotide (PT-141) is already widely available, before the FDA approval, on internet, the black market definitely learned from Viagra.

^^ That's why GHB was readily banned. It was OTC product sold as supplement. No big pharm company behind. Rohypnol wasn't banned after all so you may have a point

from what ive heard, fda is totally behind this because its most effective used sub-q, when used intranasual it takes much more to be effective, most of the population views sticking themsevles as something wrong

Yes it is, but donīt be too confident about its aphrodisiac properties.

I "misuse" it as it is very useful in the way it does openetrate the meloncorticotropin receptors and acts like an anti IGE thus attacking basophils and mastcells making them not degranulate when they contact allergenes, thus no degranulation, no toxic enzymes and cell-products being expressed, no allergy caused info-excange by chemotaxis and exacerbation = lesser allergiy.

And itīs a very good tanning aid.

In fact this means to swim, no puss runnign down his nose, despite of corticosteroids, lesser corticosteroids, more air and free sinuses as the fountains of puss are slightyl but significantly stopping to flow, nbetter asthma-comntrol.

And as a bonus, lesser appetite, inhibition of skin and lung cancer, lesser chances of emphysema as elastin is not inhibited... these new drugs are a true potion.
