Loperamide Potentiation + p-glycoprotein inhibition

SWIM is going to be performing an experiement with Loperamide, he is planning on combining the above mentioned substances ( Loperamide, Quinine, grapefruit juice, Piperazine and Cimitidine ) in hopes to to research how well they can inhibit the p-glycoproteins and allow the Loperamide to penetrate the BBB ( Blood Brain Barrier ).

SWIM is aware of the dangerous of inhibiting p-glycoprotein but is not sure how that exactly works, for how long or what the effects could be ( if any for this one time experiment ).


Any insight is appreciated, SWIM wants to be careful of course and would like to hear if anyone has tried this or similar ways of gaining any recreational use from Loperamide.

See my post at the bottom of the page there. In SWIM's case, loperamide+cimetidine resulted in what was practically death. OD lasted 24+ hours, and was only saved via multiple narcan injections, and being in the ICU for 5 days on assisted breathing. What SWIY does is your choice of coule, but in this case achieving "success" could very well be the last thing you ever accomplish in life. Just in warning in case of accidental overdose, one or even two doses or naloxone will not save oneself due to the chemicals ungodly long half-life. If not monitored by someone and given repeated injections you will just re OD again. The problem with using loperamide is there is really no way to know how much is gonna cross the BBB and become active with whatever potentiation method you are using, too many variables, what works one time may be way too much the next. There is a very fine line between nothing/mild effects, and an opiate overdose. In SWIM's opinion recreational use of loperamide is russian roulette. All in all stay safe, and be careful, I don't wish SWIM's experience on anyone.

Thanks for SWIY's opinion, SWIM was not planning on taking that massive of a dose.

He was planning on 40mg at the most with the potentiation mentioned, he doesn't think that will pose much if any.
