Bremelanotide?

Has anyone heard of Bremelanotide (a.k.a. PT-41)?

It seems to belong to a new class of drugs called melanocortids and is on track to become the first FDA-approved aphrodisiac. Currently in the last stages of preclinical trials for said approval (as of 2009).

It is not an ED drug or PDE-5 inhibitor, but seems to work by activating hypothalamic and limbic emotional structures in the brain.

It seems to work equally well on both men and women. However, there are instances of side effects, some men report a rise in blood pressure and about 1/3 of the women who tried it reported nausea.

It is available on the web via legit pharma companies, but, if interested in this, I'm sure you can find it without me giving you web-sites! You CAN, however dig up more info on this by researching Jim Pfaus, a neuroscientist out of Montreal or Mohammad Reza Safarinejad, An Iranian urologist.

Pfaus has 1300 anonymous testers in 2006 and the drug went into phase-three testing in 2007.

I haven't heard anything about it since.

Excellent post! Good to see a novel compound with such interesting potential. Great post-- good details. Keep it up!

Re: Bremelanotide?

wow, bro. an FDA approved aphrodisiac? i havent checked it out yet, but i will. for sure
do you know if this is approved as an aphrodisiac, or is this an off-label usage??(ie tegretol is for seizures, but prison MDs RX it for pain, headaches, etc. gabapentin is for seizures, but still rx for nerve pain/burning, as well as depression and headaches.)

Originally developed as (are you ready for this?) a self-tanning agent! It was repurposed when male study subjects reported a surprising side effect: erections. Fairly amazing erections! Whoda thunk it?

Re: Bremelanotide?

hmmm- according to wikip the info in this thread is somewhat off. (and while wikip is not a definitive source the info does seem to fit with what I am seeing elsewhere).

And regardless- to answer the original question it appears to be be stuck in negotiations with the FDA over the blood pressure issue.

Quote:
Bremelanotide formerly PT-141, is a compound under drug development by Palatin Technologies as a treatment for hemorrhagic shock and reperfusion injury >snip< It was originally developed for use in treating sexual dysfunction but this application was temporarily discontinued in 2008, after concerns were raised over adverse side effects of increased blood pressure. Currently, Palatin is in negotiations with the FDA to resume Human Phase 2 studies using a new subcutaneous drug delivery system that appears to have little effect on blood pressure.

Bremelanotide was developed from the peptide Melanotan II which underwent testing as a sunless tanning agent. In initial testing, Melanotan II did induce tanning but additionally caused sexual arousal and spontaneous erections as unexpected side effects in nine out of the ten original male volunteer test subjects

In studies, bremelanotide was shown to induce lordosis in an animal model and was also effective in treating sexual dysfunction in both men (erectile dysfunction or impotence) and women (sexual arousal disorder). Unlike Viagra and other related medications, it does not act upon the vascular system, but directly increases sexual desire via the nervous system

Iupac Name:(3R,6R,9S,12R,15R,23R)-15-{[(2R)-2-
Acetamidohexanoyl]amino}-9-benzyl- 6-(3-carbamimidamidopropyl)-12- (1H-imidazol-5-ylmethyl)-3-(1H-indol- 3-ylmethyl)-2,5,8,11,14,17-hexaoxo- 1,4,7,10,13,18-hexaazacyclotricosane -23-carboxylic acid

CAS number: 189691-06-3

Formula C50H68N14O10

and an article from 1999 at CNN on its parent drug Melanotan
HERE

I've also attached a so called "fact" sheet from Palatin- the pharmaceutical company that was/is developing this.

And finally- moving this to "Various drugs" forum...

Re: Bremelanotide?

Thanks! Great follow-up and good info/links for further research. My original info came from an article in a Mens magazine dated September 2009, so I wasn't too sure of its accuracy. I actually thought I UTFSE on this, but it appears I didn't. Duh! Too many things going on at once!

Re: Bremelanotide?

Hmm... I'm wondering the potential for abuse with this substance. If it works on the CNS, as opposed to just increasing erection duration or ability (such as Viagra)... it could then be used in people who don't necessarily need the substance with some degree of success, yes?

Would this be the new "ecstasy" for young couples? Or would it be the new "Rohypnol"?

... if anything at all.
